NZYFood gDNA Isolation kit has been meticulously crafted to facilitate the swift and straightforward extraction of high-purity genomic DNA from an array of diverse food and feed samples. These food matrices are inherently complex, housing an assortment of constituents such as fats, proteins, and polysaccharides, all of which can introduce challenges in DNA extraction and subsequent processing.

This kit is efficient in recovering small genomic DNA fragments, specifically those measuring less than 1 kilobase pair (kbp), from intricate food matrices, including items like ketchup and spices.

These food types typically contain minute DNA quantities and may yield degraded DNA of low quality. Considering this, we recommend the employment of primers that target short DNA fragments, typically ranging from 80 to 150 base pairs (bp). The foundation of this kit lies in the use of a highly selective silica-based matrix designed to efficiently capture nucleic acids, obviating the need for traditional, cumbersome phenol or chloroform extractions. The DNA extraction process begins with the homogenization of food/feed samples, followed by treatment with lysis buffers containing chaotropic salts, denaturing agents, and detergents. Subsequently, the lysis mixtures undergo centrifugation or filtration to eliminate contaminants and residual cellular debris.

The resulting clear supernatant is combined with binding buffer and ethanol to establish optimal conditions for DNA binding to the purposefully chosen silica membrane. This membrane possesses unique DNA-binding properties. Following this binding step, two distinct washing buffers are employed to thoroughly eliminate potential PCR inhibitors.

The extracted DNA can then be eluted using a low-salt buffer or water, rendering it ready for immediate use in subsequent applications. The quality of the eluted genomic DNA is characterized by an A260/280 ratio within the range of 1.6 to 1.9, rendering it suitable for a spectrum of enzymatic manipulations, including sequencing, real-time PCR, multiplex-PCR, genotyping, and various other molecular biology techniques. Importantly, the NZYFood gDNA Isolation kit can be effectively employed for the precise identification of GMO-DNA or animal components within food and feed samples. NZYFood gDNA Isolation kit is specifically optimized to yield up to 10 micrograms (µg) of DNA from as much as 200 milligrams (mg) of food sample, contingent upon the source of the food material being processed.
