NZYTaq 5× Optimizer Solution was developed to improve the efficacy and reduce the levels of non-specific amplifications of particular difficult PCR reactions using Taq DNA polymerase. The solution contains a proprietary range of additives and stabilizers that have proven particularly useful when attempting to amplify ancient DNA or templates contaminated with PCR inhibitors such as melanin, myoglobin or hemoglobin, for example. The addition of 5× Optimizer Solution dramatically improves yields and specificity of problematic targets in PCR amplifications using NZYTaq DNA polymerase. To test the effect of this supplementation, two different human genomic DNA sequences with GC-contents of 71.6 % (gene 1) and 65.7 % (gene 2) were amplified, in parallel, using NZYTaq DNA polymerase or NZYTaq DNA polymerase plus the optimizer solution.

Human genomic DNA used as template in the PCR reactions was isolated from human blood, which contains known PCR inhibitors. Results demonstrate a shift from non-specific products on agarose gel to the desired band when 5× Optimizer Solution was used. Lane M: NZYDNA Ladder I.
